In Raiders of the Lost Ark, Indy was supposed to have a long fight scene with the swordsman in the marketplace. This changed when Harrison Ford became ill during filming, so instead they had Indy just shoot the guy. (Which worked even better, and was pretty darn funny.)

Also, Harrison Ford was given the role of Han Solo because he happened to have been doing carpentry work on the set of Star Wars and they had him do a read through, since they didn't have anyone for the role.
